Size shifting of solid lipid nanoparticle system triggered by alkaline phosphatase for site specific mucosal drug delivery

We aim to prepare a size-shifting nanocarrier for site-targeting mucosal drug delivery that can penetrate through mucus gel layer and remain close the absorption membrane. As nanocarriers be engineered but they also back diffuse into outer regions, size shifting micron range once have reached membrane would prevent back-diffusion effect extend release over long period of time. For this purpose, we loaded solid lipid nanoparticles (SLN) with phosphate ester surfactant octadecylamine. Alkaline phosphatase (AP), bound enzyme was first time utilized as an in situ partner triggering conversion at epithelial cell surface. Having ~120 nm, SLN hydrophilic phosphate-decorated shells were shown form aggregates above Aggregates 5–8 µm formed due interparticle interactions induced by enzymatic removal after ~30 min contact isolated AP. The developed system could potential tool AP-expressing tissues like colon, lung, cervix, vagina some mucus-secreting tumors.
